Les mortalités d'huîtres observées sur les côtes françaises apparaissent principalement à proximité du sédiment et pendant la gamétogenèse. Les interactions de facteurs environnementaux sur les écosystèmes conchylicoles comme la température, les apports des bassins versants et le compartiment sédimentaire illustrent un risque de mortalité lié à différentes sources de stress (Thème 1). Les variations de la qualité de la ressource trophique pendant l'effort de gamétogenèse favorise une baisse de l'énergie disponible (Thème 2) et contraint l'organisme fragilisé à fournir un effort d'adaptation (Fent 2004). Chromosome identification is essential in oyster genomic research. Fluorescence in situ hybridization (FISH) offers new opportunities for the identification of oyster chromosomes. It has been used to locate satellite DNAs, telomeres or ribosomal DNA sequences. However, regarding chromosome identification, no study has been conducted with simple sequence repeats (SSRs). FISH was used to probe the physical organization of three particular SSRs, (GGAT)(4), (GT)(7) and (TA)(10) onto metaphase chromosomes of the Pacific oyster, Crassostrea gigas. Effects of cadmium on aneuploidy and hemocyte parameters in the Pacific oyster, Crassostrea gigas ArchiMer

Pacific oysters, Crassostrea gigas, are commonly reared in estuaries where they are exposed to anthropogenic pollution. Much research has been made on the toxicity of cadmium to aquatic organisms because the compound recurrently contaminates their environment. Our study examined the influence of cadmium on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in C. gigas at different stages of life. Adults and juveniles were exposed to two different concentrations of cadmium. Aneuploidy has previously been observed in the Pacific oyster, Crassostrea gigas, and shown to be negatively correlated with growth. Moreover, a significant impact of atrazine exposure has been described in C. gigas, and persistence of that effect has been observed between generations. Evidence of differential chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique. Pairs 1, 5, 9, and 10 are characterized by the loss of 1 chromosome. The impact of environmental factors on aneuploidy level in the oyster Crassostrea gigas was studied with experiments in controlled conditions which exposed adult and juvenile oysters to different pollutants (atrazine, a herbicide, and cadmium, a heavy metal) frequently found in Marennes-Oléron Bay. A positive correlation between atrazine concentration and aneuploidy in C. gigas was observed. Moreover, the impact of atrazine on aneuploidy level persisted in time both within and between generations, demonstrating the influence of an environmental pollution on this phenomenon. In the Pacific oyster, Crassostrea gigas hypodiploid aneuploid cells with 2n=19, 18 or 17 chromosomes have regularly been reported. There is a negative correlation between somatic aneuploidy and growth rate in oysters (Leitão et al., 2001a) as well as evidence for a genetic basis (Leitão et al., 2001b). Furthermore, non-random chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique (Leitão et al., 2001c). Impact of diuron on aneuploidy and hemocyte parameters in Pacific oyster, Crassostrea gigas ArchiMer

Diuron is a substituted urea herbicide used for agricultural and nonagricultural weed control. Its widespread use and relatively slow breakdown led us to analyze its influence on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in Pacific oysters, Crassostrea gigas. Adult oysters were subjected to two diuron concentrations (300 ng L-1 and 3 mu g L-1) for 11 weeks. Significantly higher aneuploidy level was observed in diuron-treated oysters compared with the control.
